Upscaling Human Mesenchymal Stromal Cell Production in a Novel Vertical-Wheel Bioreactor Enhances Extracellular Vesicle Secretion and Cargo Profile
This publication examines how expansion of bone marrow-derived human mesenchymal stromal cells in Vertical-Wheel® Bioreactors influences extracellular vesicle (EV) production and composition. Cells were cultured on microcarriers under controlled hydrodynamic conditions and compared with static culture systems. Bioreactor culture increased EV secretion, enhanced expression of EV biogenesis markers, and altered EV-associated microRNA and protein cargo. Proteomic and molecular analyses identified changes in pathways related to metabolism, autophagy, oxidative stress, and cellular regulation. Scale-up studies demonstrated consistent EV productivity across bioreactor volumes. The findings provide insight into how bioprocess conditions influence MSC-derived EV manufacturing and support development of scalable production strategies for cell-free therapeutics.
This guide collects and summarizes process parameters from successful experiments performed in Vertical-Wheel® bioreactors, across a variety of cell types including pluripotent stem cells, mesenchymal stem/stromal cells, exosomes and extracellular vesicles, organoids, and immune cells. Find the publication or resource most relevant to your own experiment to help with your process development.
This protocol provides detailed instructions for expanding mesenchymal stem cells on microcarriers using PBS-Mini bioreactors with Vertical-Wheel technology. The document covers the complete workflow from recovery of cryopreserved MSCs and expansion in adherent culture through inoculation, suspension culture, sampling, medium exchange, and harvest in three-dimensional bioreactor systems the PBS-Mini bioreactor. Recommended process parameters are based on internal optimization studies using multiple MSC lines and media formulations. Guidance is provided for microcarrier preparation, cell…
This user manual provides instructions for setting up, calibrating, and operating the PBS 0.5 Mini Vessel with a 60 µm perfusion filter. It covers required materials, pump configuration, vessel preparation, tubing installation, medium exchange, perfusion initiation, and routine operation. Step-by-step procedures address feed and waste management, flow-rate verification, troubleshooting, and maintenance. The manual serves as a practical reference for implementing continuous medium exchange and perfusion workflows within the PBS-MINI platform while maintaining controlled, sterile operation.
